Cloned(Commentary)

EC Number Cloned (Comment) Organism
2.5.1.B43 gene bchG, recombinant expression of wild-type and mutant enzymes in Escherichia coli strain BL21(DE3) Cereibacter sphaeroides
2.5.1.62 gene chlG, recombinant expression of wild-type and mutant enzymes in Escherichia coli strain BL21(DE3), functional recombinant expression of mutant I44F in Rhodobacter sphaeroides photosynthesis-deficient mutant strain BG1 that lacks a functional bchG gene encoding for bacteriochlorophyll synthase, EC 2.5.1.133 Synechocystis sp. PCC 6803

Protein Variants

EC Number Protein Variants Comment Organism
2.5.1.B43 F28I naturally occuring mutation, the mutant shows chlorophyll synthase, EC 2.5.1.62, ChlG, activity Cereibacter sphaeroides
2.5.1.B43 additional information a BchG mutant Rhodobacter sphaeroides BG1 strain is photosynthetically incompetent. Expression of gene chlG from Synechocystis sp. PCC 6803, encoding a chlorophyll synthase I44F mutant, EC 2.5.1.62, rescues the photoheterotrophical growth of the mutant cells. The Synechocystis mutated enzyme ChlGI44F shows phytyl-dependent bacteriohchlorophyll a synthase activity Cereibacter sphaeroides
2.5.1.62 I44F naturyll occuring mutant, that also shows bacteriochlorophyl syntase activity, EC 2.5.1.133 Synechocystis sp. PCC 6803

General Information

EC Number General Information Comment Organism
2.5.1.B43 evolution the predicted sequence of ChlG of Synechocystis sp. PCC 6803 is considerably similar (35% identity) to that of Rhodobacter sphaeroides BchG, each enzyme has a high level of substrate specificity to distinguish its own substrate from the other. But the BchG activity of Synechocystis chlorophyll synthase mutant ChlGI44F and the ChlG activity of bacteriochlorophyll synthase mutant BchGF28I suggest that ChlG/EC 2.5.1.62 and BchG/EC 2.5.1.B43 are evolutionarily related enzymes, cf. EC 2.5.1.133 Cereibacter sphaeroides
2.5.1.B43 additional information residue F28 is important for the substrate specificities of the enzyme Cereibacter sphaeroides
2.5.1.62 evolution the predicted sequence of ChlG of Synechocystis sp.
