physiological function proprotein convertase PC1/3 regulates the endosomal toll-like receptor TLR9. Under CpG oligodesoxynucleotide challenge, PC1/3 traffics rapidly to colocalize with TLR9 in CpG oligodesoxynucleotide-containing endosomes with acidic pH. In PC1/3 knockdown macrophages, compartmentalization of TLR9 is altered and TLR9 clustered in multivesicular bodies. This clustering of TLR9 in multivesicular bodies dampens the anti-inflammatory STAT3 signalling pathway while it promotes the pro-inflammatory NF-kappaB pathway Mus musculus
