a Solanum tuberosum cDNA fragment gamma-TMT is introduced into intron-spliced hairpin RNA, RNAi, silencing construct driven by the constitutive cauliflower mosaic virus 35S promoter. RNAi knockdown of gamma-TMT results in an up to 95% reduction of alpha-tocopherol in leaves of transgenic plants, which is almost quantitatively compensated for by an increase in gamma-tocopherol. gamma-TMT-silenced plants show an improved performance when challenged with sorbitol or methyl viologen, but salt tolerance of gamma-TMT transgenic plants is strongly decreased, compared to the wild-type plants, phenotype, overview |
