Protein Variants

EC Number Protein Variants Comment Organism

2.1.1.379 H375F the mutant sustains a 0.5% loss in Arg275 methylation as compared to the wild type enzyme Methanococcus maripaludis
2.1.1.379 H375F/DELTAN371 the mutant sustains a 42.5% loss in Arg275 methylation as compared to the wild type enzyme Methanococcus maripaludis
2.1.1.379 H375G the mutant sustains a 0.8% loss in Arg275 methylation as compared to the wild type enzyme Methanococcus maripaludis
2.1.1.379 N212A/D213A the mutant sustains a 35% loss in Arg275 methylation as compared to the wild type enzyme Methanococcus maripaludis
2.8.4.1 additional information markerless deletion of methanogenesis marker protein 10 (Mmp10), encoded by gene mmpX, can play an important role in methanogenesis. To systematically examine its physiological role, mmpX (locus tag MMP1554), the deletion of gene encoding Mmp10 in Methanococcus maripaludis results in the complete loss of the 5-C-(S)-methylarginine PTM of residue 275 in the McrA subunit. When the DELTAmmpX mutant is complemented with the wild-type gene expressed by either a strong or a weak promoter, methylation is fully restored. Compared to the parental strain, maximal rates of methane formation by whole cells are reduced by 40 to 60% in the DELTAmmpX mutant. The reduction in activity is fully reversed by the complement with the strong promoter. Site-directed mutagenesis of mmpX result in a differential loss of arginine methylation among the mutants in vivo. Construction of diverse mutant strains from parental strain S0001, and phenotypes, overview Methanococcus maripaludis

Posttranslational Modification

EC Number Posttranslational Modification Comment Organism
2.8.4.1 alkylation the posttranslational methylation of arginine in methyl coenzyme M reductase has a profound impact on both methanogenesis and growth of Methanococcus maripaludis. Protein Mmp10 is necessary for arginine methylation. Replacement of the only conserved His residue in the C-terminal half of Mmp10 with either G or F results in a loss of less than 1% of Arg275 methylation Methanococcus maripaludis
2.8.4.1 additional information the McrA subunit possesses up to five highly conserved but not universal posttranslational modifications (PTMs) at its active site, including 1-N-methyl-His261, 5-C-(S)-methyl-Arg275, 2-(S)-methyl-Gln403, and thio-Gly448, which are all found within the McrA subunit Methanococcus maripaludis
