NaCl in the presence of a low amount of NaCl (10 mM), TopR2 is able to produce DNA with a very high number of positive superturns. At 100 mM NaCl TopR2 produces the highest amount of this highly positively supercoiled DNA. Further increase in NaCl concentration until 400 mM does not provide a further higher amount of positively supercoiled DNA. At 600 mM NaCl, the presence of some positively supercoiled topoisomers is still observed but no more at 800 mM Saccharolobus solfataricus
