Plasmids with different promoters and copy numbers are important for the expression of trehalose synthase genes in Escherichia coli. The trehalose synthase from Thermus thermophilus is best expressed from plasmid pET-22b showing high activity in trehalose production with 1.996 U/mg protein, co-expression of molecular chaperones sigma32, GroEL, GroES, DnaK and DnaJ greatly increasing the solubility of the trehalose synthase protein Thermus thermophilus
