Cloned(Commentary)

Cloned (Comment) Organism
a vaccinia virus expression system is used for the production of recombinant human cathepsin D in human cell lines MCF7 (breast cancer), HepG2 (hepatocarcinoma) and HeLa (cervix cancer) and non-human cell line BSC1 (simian kidney epithelial). For generation of a recombinant vaccinia virus the full-length cathepsin D cDNA comprising the ATG codon is inserted downstream of a synthetic vaccinia late promoter into the vector pMJ601 which also contains the Escherichia coli LacZ gene. This expression cassette is flanked by segments of the vaccinia thymidine kinase (TK) gene that allows homologous recombination with the wild-type virus Homo sapiens

Posttranslational Modification

Posttranslational Modification Comment Organism
glycoprotein proCDrec is largely secreted into the culture medium, although it contains high-mannose oligosaccharides with uncovered mannose-6-phosphate residues Homo sapiens
proteolytic modification intracellular proCDrec is processed into the 48 kDa intermediate single-chain and the 31 plus 13 kDa double-chain form. Processing is slower than in normal cells Homo sapiens
