Activating Compound

Activating Compound Comment Organism Structure
additional information pehA transcription is greatly induced by polygalacturonic acid and positively regulated by Clp (cAMP receptor protein-like protein) and RpfF (an enoyl-CoA hydratase homologue which is required for the synthesis of cis-11-methyl-2-dodecenoic acid, a low-molecular-mass diffusible signal factor) Xanthomonas campestris pv. campestris

Application

Application Comment Organism
additional information PehA is the major but not the sole polygalacturonase, it plays a minor role in Xanthomonas campestris virulence Xanthomonas campestris pv. campestris

Cloned(Commentary)

Cloned (Comment) Organism
1620 bp DNA fragment encompassing the upstream 218 bp plus the entire coding region of the Xc17 pehA into the PstI-XbaI sites of the broad-host-range vector pRK415. 1376 bp NdeI-XhoI fragment containing the Xc17 pehA gene cloned in the Escherichia coli vector pOK12, giving pOKpeh. A Gmr cartridge from pUCGM inserted into the unique HincII site within the pOKpeh insert. Resultant plasmid, pOKpehG, electroporated into strain Xc17 allowing for double crossover. PpehA-lacZ transcriptional fusion constructs generated by cloning PCR fragments into the broad-host-range promoter-probing vector pFY13-9, which uses lacZ as the reporter Xanthomonas campestris pv. campestris

Protein Variants

Protein Variants Comment Organism
additional information in mutant MH172 with cloned pehA, MH172(pRKpeh), wild-type-level polygalacturonase activity is restored Xanthomonas campestris pv. campestris

Inhibitors

Inhibitors Comment Organism Structure
additional information expression of pehA is subject to catabolite repression, which is independent of Clp and RpfF, and it is repressed under conditions of oxygen limitation or nitrogen starvation Xanthomonas campestris pv. campestris
