Cloned (Comment) Organism
gene CKA4 or At2g23070, recombinant overexpression of GFP-tagged subunit CK2 alpha4 in Arabidopsis thaliana via Agrobacterium tumefaciens mediated transformation Arabidopsis thaliana

Protein Variants

Protein Variants Comment Organism
additional information enzyme knockout by construction of two independent CK2 alpha4 RNAi lines in the CK2 alpha triple mutant background, phenotype, overview. The RNAi lines in the wild-type background show 70-80% relative kinase activity while the relative kinase activity in the RNAi lines in CK2 alpha triple mutant background is reduced to 40-50%. Although the RNAi lines show up to 70-75% reduction inthe transcript level it does not correlate with the reduction seen in the relative kinase activity of the plant extracts. It islikely that the RNAi lines with severe reduction in kinase activity may not be viable and hence are not obtained from the screen Arabidopsis thaliana

Localization

Localization Comment Organism GeneOntology No. Textmining
chloroplast GFP-tagged CK2 alpha4 localizes to the chloroplast in transgenic Arabidopsis seedlings, consistent with the presence of a chloroplast localization signal at the N-terminus of CK2 alpha4 subunit. Subunit CK2 alpha4 also contains the internal conserved nuclear localization signal, NLS Arabidopsis thaliana 9507
-
nucleus in Arabidopsis thaliana, both alpha and beta subunits are redundantly encoded by four genes each and are ubiquitously expressed spatially and developmentally. All the CK2 alpha subunits except alpha4 are nuclear localized. Subunit CK2 alpha4 has an N-terminal chloroplast localization signal and also contains the internal conserved nuclear localization signal, NLS Arabidopsis thaliana 5634

General Information

General Information Comment Organism
malfunction both CK2 alpha4 single knockout and CK2 alpha3alpha4 double knockout mutants are hyposensitive to abscisic acid induced block of seed germination Arabidopsis thaliana
physiological function three of the four CK2 alpha subunit types existing in Arabidopsis act in a functionally redundant manner to regulate various developmental pathways. The fourth alpha subunit in Arabidopsis thaliana, CK2 alpha4, also functions redundantly in regulating abscisic acid response, lateral root formation and flowering time, functionally overlapping role for the CK2 alpha4 subunit in regulating various developmental processes in plants. CK2 alpha4 positively regulates the block of seed germination inpresence of abscisic acid. Arabidopsis, the nuclear CK2 alpha subunits contribute synergistically to the block of seedgermination in response to hormone abscisic acid and salt in aconcentration dependent manner Arabidopsis thaliana
