E204G mutation to the corresponding residue of Coptis japonica coclaurine N-methyltransferase. The mutation has similarly modest (negative) effects on Km and kcat for (S)-N-methylcoclaurine, but with respect to (S)-coclaurine has a substantial (positive) effect only on kcat Papaver somniferum
G204E the mutation results in an enzyme with reduced canonical reticuline N-methyltransferase activity and increased canonical coclaurine N-methyltransferase activity. The mutant enzyme shows activity with (S)-coclaurine, which is modestly better than with (S)-N-methylcoclaurine but worse than with (S)-reticuline. A G204E substitution has no appreciable effect on Km for (S)-coclaurine but increases Km for (S)-N-methylcoclaurine approximately 4fold. kcat for (S)-coclaurine is 10fold greater while that for (S)-N-methylcoclaurine decreases 4fold. The G204E substitution has similarly modest (negative) effects on Km and kcat for (S)-N-methylcoclaurine, but with respect to (S)-coclaurine has a substantial (positive) effect only on kcat Papaver somniferum
