The enzyme, charactrized from the bacterium Bacillus subtilis, catalyses a similar tRNA modification to that performed by EC 2.3.1.193, tRNAMet cytidine acetyltransferase. However, unlike that enzyme, which uses acetyl-CoA as the acetyl donor, this enzyme activates an acetate ion to form acetyladenylate and then catalyses the acetylation through a mechanism similar to tRNA aminoacylation.
The expected taxonomic range for this enzyme is: Bacillus subtilis

the enzyme is responsible for formation of N4-acetylcytidine34 at the anticodon first position (position 34) of bacterial elongator tRNAMet. Modification of tRNA anticodons plays a critical role in ensuring accurate translation. ac4C34 modification of tRNAeMet prevents misdecoding of AUA cooperatively with L34 modification of tRNAIle2
