Sulfation takes place at the 6-position of galactosyl and N-acetylglucosaminyl residues in keratan, a proteoglycan. Not identical with EC 2.8.2.5 (chondroitin 4-sulfotransferase), EC 2.8.2.6 (choline sulfotransferase) or EC 2.8.2.17 (chondroitin 6-sulfotransferase).

KSST competes with fucosyltransferase-VII for the same acceptor substrate and downregulates the synthesis of L-selectin ligand by inhibiting alpha1,3-fucosylation, sulfated oligosaccharide, e.g. 6'-sulfated galactose residues, increase in FucT-VII-deficient mutant mice, structure of mono-sulfated O-glycans, detailed overview

enzyme GlcNAc-6-sulfated KS is recognized by the R-10G anti-KS antibody, of which the minimum epitope structure is Galbeta1-4GlcNAc(6S)beta1-3Galbeta1-4GlcNAc(6S), and distributes diffusely in neuropils and presents densely in close proximity to the perineuronal region of the perineuronal net (PNN)-positive neurons in the adult visual cortex
the enzyme is required for the generation of Siaalpha2->3[6S]beta->4[6S]GlcNAc or a closely related structure in lymph node high endothelial venules. The enzyme does does not contribute to L-selectin ligand expression or lymphocyte homing
keratan sulfate (KS) is a carbohydrate side chain covalently attached to extracellular proteoglycans. KS is composed of disaccharide units of 6-sulfated N-acetylglucosamine (GlcNAc) and galactose. GlcNAc6ST3 is a major sulfotransferase for the R-10G-positive KS in the adult mouse brain. GlcNAc-6-O-sulfotransferase (GlcNAc6ST) 1 encoded by Chst5 is an enzyme necessary for the synthesis of GlcNAc-6-sulfated KS chains which are required for neuronal plasticity in the visual cortex of the mouse brain during the critical period, but not in adulthood. GlcNAc-6-sulfated KS is recognized by the R-10G anti-KS antibody, of which the minimum epitope structure is Galbeta1-4GlcNAc(6S)beta1-3Galbeta1-4GlcNAc(6S), and distributes diffusely in neuropils and presents densely in close proximity to the perineuronal region of the perineuronal net (PNN)-positive neurons in the adult visual cortex. GlcNAc6ST3, which has been discovered as an intestinal GlcNAc6ST encoded by Chst5, is a major brain KS sulfotransferase expressed in oligodendrocytes in adulthood. An isoform of the protein-tyrosine phosphatase PTPRZ is also a R-10G-reactive KS proteoglycan. GlcNAc6ST3 may play a role in synthesis of a component of PNN in the adult brain, and the KS-modified isoform of PTPRZ encoded by Ptprz1 might be an extracellular molecule associated with PNNs
overexpression of KSST suppresses the expression of sialyl Lewis X on CHO cells engineered to express sialyl Lewis X, KSST expression in those cells suppresses lymphocyte rolling compared with mock-transfected CHO cells expressing 6-sulfo sialyl Lewis X, overview
Significant decrease in alpha1,3-linked fucose in association with increase in 6-sulfated N-acetylglucosamine in peripheral lymph node addressin of FucT-VII-deficient mice exhibiting diminished lymphocyte homing
