The assay uses the cohesive ends of restriction endonuclease digested lambda-DNA as substrate. Separation of ligated from unligated cohesive end-containing fragments via agarose gel electrophoresis, followed by digital image analysis employing a normalization procedure to correct for artifacts of gel electrophoresis, allows the accurate determination of unit ligase activity
