thymine-mismatched double-stranded DNA + H2O the recombinant enzyme shows a significant preference for G/T mispairs in a CpG context. The enzyme is also capable of processing mismatches between thymine and 6-O-methylguanine, whereby it generates an apyrimidinic site opposite the modified guanine. TDG preferentially addresses 6-O-methyl G/T mispairs when the neighboring base 5' to the 6-O-methyl G is a C rather than a G.
