Crystallization (Comment) | Organism |
---|---|
two different crystal forms: higher density and lower density determined at 1.40 A and 1.26 A resolution, respectively. The crystal structures reveal an unusual packing mode, clearly visible in the LD structure, caused by the presence of the C-terminal His6 tag, which extends from the compact fold of the enzyme molecule and docks in the catalytic cleft of a neighbouring molecule. In this way, an infinite chain of His-tag-linked protein molecules is formed along the c direction. Purified recombinant His-tagged enzyme by hanging drop vapour diffusion method, mixing of 0.0015 ml of 8 mg/ml protein in 20 mM Tris-HCl, pH 8.0, with 0.0015 ml of reservoir solution containing 0.1 M sodium acetate, pH 4.6, 0.2 M ammonium acetate, 25-30% PEG 4000, 21°C, 2 days, X-ray diffraction structure determination and analysis, modelling | Solanum tuberosum |
