Cells are harvested by centrifugation, and the cell pellet is suspended in HEPES buffer containing 2 mM DTT and 0.2 mM phenylmethanesulfonyl fluoride. The cells are lysed in a French press and the lysate is centrifuged. The supernatant is loaded onto a DEAE Sepharose Fast Flow column equilibrated with HEPES buffer. A linear gradient of KCl in 50 mM Hepes is used to elute the protein. Desired fractions are pooled, concentrated and subjected to ammonium sulfate fractionation. The 40-60% ammonium sulfate fraction is centrifuged, the protein pellet is dissolved, loaded onto a size exclusion column and eluted with 10 mM HEPES containing 0.15 M KCl.
